A Gain-of-Function Mutation in DHT Synthesis in Castration-Resistant Prostate Cancer
Scientists showed that castration-resistant prostate cancer sometimes expresses a gain-of-stability mutation that leads to a gain-of-function in 3ß-hydroxysteroid dehydrogenase type 1, which catalyzes the initial rate-limiting step in conversion of the adrenal-derived steroid dehydroepiandrosterone to dihydrotestosterone (DHT). Targeting the Androgen Receptor with siRNA Promotes Prostate Cancer Metastasis through Enhanced Macrophage Recruitment via CCL2/CCR2-Induced STAT3 Activation

The Twist Box Domain Is Required for Twist1-Induced Prostate Cancer Metastasis
Investigators examined a transactivation mutant, Twist1-F191G, in prostate cancer cells using in vitro assays which mimic various stages of metastasis. Twist1 overexpression led to elevated cytoskeletal stiffness and cell traction forces at the migratory edge of cells based on biophysical single-cell measurements. [Mol Cancer Res] Abstract

The Anticancer Effect of Fucoidan in PC-3 Prostate Cancer Cells
Fucoidan induced the apoptosis of PC-3 cells by activating both intrinsic and extrinsic pathways. The induction of apoptosis was accompanied by the activation of extracellular signal-regulated kinase mitogen-activated protein kinase (ERK1/2 MAPK) and the inactivation of p38 MAPK and phosphatidylinositol 3-kinase/Akt. Inhibition of SIRT6 in Prostate Cancer Reduces Cell Viability and Increases Sensitivity to Chemotherapeutics
Knockdown of SIRT6 in human prostate cancer cells led to sub-G1 phase arrest of cell cycle, increased apoptosis, elevated DNA damage level and decrease in BCL2 gene expression. [Protein Cell] Abstract

Attenuation of Nucleoside and Anti-Cancer Nucleoside Analog Drug Uptake in Prostate Cancer Cells by Cimicifuga racemosa Extract BNO-1055
Scientists investigated the mechanisms underlying the anti-proliferative effects of the ethanolic Cimicifuga racemosa extract BNO-1055 on prostate cells and evaluate its therapeutic potential. BNO-1055 dose-dependently attenuated cellular uptake and incorporation of thymidine and BrdU and significantly inhibited cell growth after long-time exposure. [Phytomedicine] Abstract

Loss of P53 Facilitates Invasion and Metastasis of Prostate Cancer Cells
Researchers found si-P53 promotes the invasion and metastasis in prostate cancer cells, increases the expression and activity of MMP-2/9 and MMP-14. [Mol Cell Biochem] Abstract

miR-205 Is Frequently Downregulated in Prostate Cancer and Acts as a Tumor Suppressor by Inhibiting Tumor Growth
Functional analyses showed that both the overexpression of microRNA-205 (miR-205) and the knockdown of c-SRC in prostate cancer cell lines could inhibit cell growth, colony formation, migration, invasion and the cell cycle as well as induce cell apoptosis in vitro. [Asian J Androl] Full Article

The miRNA let-7a1 Inhibits the Expression of Insulin-Like Growth Factor 1 Receptor (IGF1R) in Prostate Cancer PC-3 Cells

A new survey suggests that men may not always know just what the doctor has ordered when it comes to PSA screening. In a survey of 1,539 men, 80 percent were unaware that the U.S. Preventive Services Task Force has recommended against screening for prostate cancer in healthy men.
